Replicating, Revising and Reforming: Unpicking the Apparent Nanoparticle Endosomal Escape Paradox

Over the past decade there has been an exponential increase in the number of research papers utlising nanoparticles for biological applications such as intracellular sensing [1, 2], theranostics [3-5] and more recently drug delivery and precision medicine [6, 7]. Despite the success stories, there is a disconnect regarding current dogma on issues such as nanoparticle uptake and trafficking, nanoparticle delivery via the enhanced permeability and retention (EPR) effect, and endosomal escape. Critical re-evaluation of these concepts both conceptually and experimentally is needed for continued advancement in the field.
 
For this preregistration, Said et al. (2024) focus on nanoparticle intracellular trafficking, specifically endosomal escape [8]. The current consensus in the literature is that nanoparticles enter cells via endocytosis [9, 10] but reportedly just 1-2% of nanoparticles/ nanoparticle probes escape endosomes and enter the cytoplasm [11-13]. There is therefore an apparent paradox over how sensing nanoparticles can detect their intended targets in the cytoplasm if they are trapped within the cell endosomes. To address this fundamental issue of nanoparticle endosomal escape, Lévy and coworkers are carrying out replication studies to thoroughly and transparently replicate the most influential papers in the field of nanoparticle sensing. The aim of these replication studies is twofold: to establish a robust methodology to study endosomal escape of nanoparticles, and to encourage discussions, transparency and a step-change in the field.  
 
Replication of “Carbon-Dot-Based Dual-Emission Nanohybrid Produces a Ratiometric Fluorescent Sensor for In Vivo Imaging of Cellular Copper Ions”
 
For this replication study, the authors classified papers on the topic of nanoparticle sensing and subsequently ranked them by number of citations.  Based on this evaluation they selected a paper by Zhu and colleagues [14] entitled “Carbon-Dot-Based Dual-Emission Nanohybrid Produces a Ratiometric Fluorescent Sensor for In Vivo Imaging of Cellular Copper Ions” for their seminal replication study.  To determine the reproducibility of the results from Zhu et al., the authors aim to establish the proportion of endosomal escape of the nanoparticles, and to examine the data in a biological context relevant to the application of the probe.
 
Beyond Replication
 
The authors plan to replicate the exact conditions reported in the materials and methods section of the selected paper such as nanoparticle probe synthesis of CdSe@C-TPEA nanoparticles, assessment of particle size, stability and reactivity and effect on cells (TEM, pH experiments, fluorescent responsivity to metal ions and cell viability). In addition, Said et al., plan to include further experimental characterisation to complement the existing study by Zhu and colleagues. They will incorporate additional controls and methodology to determine the intracellular location of nanoparticle probes in cells including: quantifying excess fluorescence in the culture medium, live cell imaging analysis, immunofluorescence with endosomal and lysosomal markers, and electron microscopy of cell sections. The authors will also include supplementary viability studies to assess the impact of the nanoparticles on HeLa cells as well as an additional biologically relevant cell line (for use in conjunction with the HeLa cells as per the original paper).
 
The Stage 1 manuscript underwent two rounds of thorough in-depth review. After considering the detailed responses to the reviewers' comments, the recommenders  determined that the manuscript met the Stage 1 criteria and awarded in-principle acceptance (IPA).  
 
The authors have thoughtfully considered their experimental approach to the replication study, whilst acknowledging any potential limitations. Given that conducting such a replication study is novel in the field of Nanotechnology and there is currently no ‘gold standard’ approach in doing so, the authors have showed thoughtful regard of statistical analysis and unbiased methodology where possible.
 
Based on current information, this study is the first use of preregistration via Peer Community in Registered Reports and the first formalised replication study in Nanotechnology for Biosciences. The outcomes of this of this study will be significant both scientifically and in the wider context in discussion of the scientific method.

Personality traits predict perception of pandemic risk and compliance with infection control measures

Associations between personality traits, compliance and risk perception in Norwegian adults during COVID-19

This study aims to examine the associations between personality traits, using the big-5 personality model (Costa & McCrae, 1992), and compliance with pandemic measures (e.g. handwashing), and perceived risk of the pandemic to the individual, in a representative sample of the adult Norwegian population.
 
Several previous studies have examined associations between personality and adherence to medical advice using measures administered at the same point in time the during COVID-19 pandemic (e.g. Götz et al., 2021), however the dataset the current investigators plan to use assessed personality one year prior to the pandemic (May/June 2019) and the risk and compliance measures four months into the pandemic (August/September 2020). 
 
Here, Sætrevik et al. (2024) make a number of hypotheses, including that conscientiousness, agreeableness, openness and neuroticism will be positively associated with compliance; extraversion will be negatively associated with compliance; extraversion and openness will be negatively associated with perceived risk; and finally, neuroticism will be positively associated with perceived risk. They plan to test their hypotheses using linear regression analyses.
 
Importantly, given the topic, the data for this study already exists and some response distributions of the compliance and risk measures have been examined. However, the authors have yet to link the data on personality to the data on compliance and risk, which is the focus of their hypotheses.  
 
The findings from this study will help us to understand the personality characteristics associated with infection control measures and individual risk perceptions of viral illnesses during a pandemic.

Dose-response of tDCS effects on motor learning and cortical excitability: a preregistered study

How stimulation intensity affects motor learning

In neurostimulation research, the parameters of a stimulation protocol crucially impact on the effects of the stimulation. Transcranial direct current stimulation (tDCS) is a neurostimulation technique that typically uses current intensities about 1-2 mA in human research to modulate motor and cognitive behavior. The current sham-controlled study by Hsu et al. (2024) applies current intensities not only of 2 mA but also of 4 mA and 6 mA and thus extends our understanding of stimulation parameters while ethical standards are preserved.
 
The influence of tDCS over the primary motor cortex will be evaluated for neural plasticity during motor learning. Stimulation effects will be tested not only behaviorally but also physiologically by motor evoked potentials elicited by transcranial magnetic stimulation (TMS). The presented pilot data are promising and underline the feasibility of the proposed research design. The study will contribute to tDCS research by uncovering reasons for controversial findings and thus increase reproducibility.

Revisiting the Psychology of Waste: Replication and extensions Registered Report of Arkes (1996)

When do perceptions of wastefulness affect how people make choices?

How do perceptions of wastefulness affect how people make choices? In an influential set of studies examining different conceptions of wasteful behavior (overspending, underutilization, and sunk costs), Arkes (1996) found a systematic aversion to wastefulness in decision making, even when choosing to avoid wastefulness has no economic value or works against personal interest. While these findings have been influential in basic and applied research, there have been no attempts to directly replicate the results. Moreover, the original study has several methodological limitations, including the use of relatively small samples and gaps in statistical analysis and reporting.
 
In this Stage 1 manuscript, Zhu and Feldman (2024) propose to conduct a high-powered replication of Arkes (1996) using an online sample of participants. The authors will incorporate several extensions to improve methodological rigor relative to the original article, including added comprehension checks, checks of the wastefulness manipulations, a within-subjects design, and a quantitative analysis of participants’ self-reported motivations for their choices. The results of the study will provide insight into the robustness of the original findings, while also better distinguishing wastefulness aversion from other potential reasons behind participants' decisions.

Test-Retest Reliability of the STRAQ-1: A Registered Report

We may not be measuring physical closeness in interpersonal relationships as reliably as we think

Attachment and interpersonal relationships are a major subject of research and clinical work in psychology. There are, accordingly, a proliferation of measurement instruments to tap into these broad constructs. The emphasis in these measures tends to be on the emotional dimensions of the relationships—how people feel about their partners and the support that they receive. However, that is not all there is to relationship quality. Increasing attention has been paid to the physical and physiological aspects of relationships, but there are few psychometrically sound measures available to assess these dimensions.
 
In the current study, Dujols et al. (2024) assessed the psychometric properties of the Social Thermoregulation and Risk Avoidance Questionnaire (STRAQ-1), a measure of physical relationships that targets social thermoregulation, or how physical proximity is used to promote warmth and closeness. The project consists of a thorough assessment of the measure’s reliability over time—that is, the degree to which the measure assesses the construct similarly across administrations, in a sample of 183 French university students.
 
The authors assessed the longitudinal measurement invariance and test-restest reliability of the STRAQ-1. Longitudinal measurement invariance across two time points was only found for two of the four subscales. Similarly, test-retest reliability varied by subscale, ranging from poor to good. Taken together, the study suggests caution in using the STRAQ-1 scale as a reliable measure of physical relationships. The study highlights the need for continued assessment of the reliability of widely used measures, particularly reliability over time, and serves as a model for a rigorous analytic approach for doing so.

Unveiling the Positivity Bias on Social Media: A Registered Experimental Study On Facebook, Instagram, And X

Both research and public debates around social media use tend to involve a premise of positivity bias, which refers to presenting one’s life in an overly positive light by various different means. This premise contributes to multiple potentially important follow-up hypotheses, such as the fear of missing out and low self-image effects, due to repeated consumption of positive social media content (e.g., Bayer et al. 2020, for a review). The positivity bias of social media use, itself, has received limited research attention, however. 
 
In the present study, Masciantonio and colleagues (2024) will test positivity bias in the context of three social media platforms: Facebook, Instagram, and X. The experiment involves recruiting participants into platform-specific user groups and crafting posts to be shared with friends as well as respective social media audiences. If positivity bias manifests in this context, the social media posts should introduce more positive valence in comparison to offline sharing—and if the platforms differ in their encouragement of positivity bias, they should introduce significant between-platform differences in valence.
 
The Stage 1 plan was reviewed by four independent experts representing relevant areas of methodological and topic expertise. Three reviewers proceeded throughout three rounds of review, after which the study was considered having met all Stage 1 criteria and the recommender granted in-principle acceptance.

The importance of conceptual knowledge when becoming familiar with faces during naturalistic viewing

A registered test of the role of contextual information in perceptual learning of faces

When we familiarise with new faces over repeated exposures, it is generally in situations that have meaning for us. Seeing a face more often tends to go along with learning more about the person, and their likely contexts and actions. In this Registered Report, Noad and Andrews (2024) tested whether meaningful context during exposure improves the consolidation of faces into long-term memory. Participants were shown video clips from the TV series Life on Mars, either in their original chronological sequence, which provides meaningful context, or in a scrambled sequence. It was expected that the original sequence would provide a better conceptual understanding, and this was confirmed by free recall and structured question tests. Face recognition memory was tested with images of the actor from the original clips (‘in show’) and the same actor from another show (‘out-of-show’), to test whether memory was modulated by the similarity of appearance to that at encoding. Face recognition was tested immediately after exposure and after four weeks, to allow time for consolidation. As expected, recognition memory was better for participants in the meaningful context condition, and for in-show faces. However, meaningful context did not lead to less forgetting of the faces at the follow up test, even for in-show faces, which did not support the original predictions. An exploratory analysis found that a metric of overlap between pairs of participants’ conceptual understanding was related to overlap in the set of faces they recognised. This relationship was stronger after four weeks, which suggests increased interaction of conceptual knowledge and face recognition after consolidation.

Understanding probability assessments with partitioned framing

Decision-making based on limited information is a common occurrence. Whether it is the possibility of a cheaper product elsewhere or the unknown qualifications of election candidates, people are regularly forced to make a decision under ignorance or uncertainty. In such situations, information about certain events is unavailable or too costly to acquire and people rely on subjective probability allocation to guide decision-making processes. This allocation seems to result in what is known as ignorance priors, i.e., decision-makers assigning equal probabilities to each possible outcome within a given set. How events are grouped or partitioned is often subjective and may influence probability judgments and subsequent decisions. In such cases, the way the choices within a choice set are presented may shape the perceived likelihood of different outcomes. Understanding the impact of partitioning on probability estimation is crucial for both psychological and economic theories of judgment and decision.
 
The question of evaluating probabilities under uncertainty has received much attention in the psychology and economics literature over the past decades given the wide range of possible applications. In the current work, Ding and Feldman (2024) seek to replicate one of the foundational works on the topic: Fox and Rottenstreich (2003). In the original work, the authors provided exploratory evidence indicating that the framing of a situation affects the way individuals perceive probabilities of possible outcomes. They showed that people assigned uniform probabilities to sets of events described in a problem, such that the way the events are described partly determines people’s partitioning of those events and evaluations of the probabilities of the possible outcomes. Additionally, this partitioned framing affected judgments both under conditions of ignorance (where individuals have no information and rely solely on uniform probability assignments) and uncertainty (where individuals have some information but still rely on heuristics influenced by partitioning). This suggests that priors resulting from the inference of available evidence are sometimes partly contaminated by partitioning bias, affecting both uninformed and partially informed decision-making processes. As a consequence, the partitioning of events into different subsets might lead to varying evaluations of a single situation, resulting in inconsistencies and poorly calibrated probability assessments.
 
Ding and Feldman (2024) aim to replicate Studies 1a, 1b, 3, and 4 from Fox and Rottenstreich (2003). Their close replication will rely on original data (US participants, Prolific, N=600, not collected yet) with a large statistical power (>95%). Their replication aims to examine whether the partitioned framing affects prior formation under ignorance (Studies 1a, 1b, and 4) and uncertainty (Study 3). In addition, the authors propose an extension examining estimations of alternative event(s) contrasting estimations of the probabilities of events happening versus of events not happening.
 
The Stage 1 manuscript was evaluated by two external reviewers and the recommender. Based on detailed responses to the reviewers' and the recommender’s comments, the recommender judged that the manuscript met the Stage 1 criteria and therefore awarded in-principle acceptance (IPA).

Is it Worth the Hustle? A Multi-Country Replication of the Effort Moralization Effect and an Extension to Generational Differences in the Appreciation of Effort

Are people who exert more effort in a task seen as more moral?

This study seeks to understand cultural and age differences in the effort moralization effect, a phenomenon in which people who put more effort into a task are considered more moral, regardless of the quality or the morality associated with the task. This is shown in common phrases such as the “great resignation” or “quiet quitting”, which are mostly used against younger members of the population, in particular generation Z.
 
Tissot and Roth (2024) propose to conduct a replication of a study from Celniker et al. (2023) which found evidence for this effect, with new samples from Mexico and Germany to test potential cultural differences. They will also test the effect of age on the effort moralization effect. Therefore, the study will be a quantitative analysis.
 
The authors included an adequate power analysis, alternatives for non-supported hypotheses, and filtering to ensure a high quality of data collection. They already provided an R script and dummy data to ensure the quality of the analysis.
 
The Stage 1 manuscript was evaluated over three rounds of in-depth review. Based on ​detailed responses to reviewers’ and the recommender’s comments, the recommender judged that the manuscript met the Stage 1 criteria and therefore awarded in-principle acceptance.​​​

Recent studies using an EEG frequency tagging approach have reported modulations of alpha, beta and theta bands at the stimulation frequency during nociceptive/painful thermal stimulation compared to non-nociceptive/non-painful vibrotactile stimulation. Prior expectations of the intensity of upcoming painful stimuli are known to strongly modulate the subjective experience of those stimuli. Thus, modulating the expectation of pain should result in a change in the modulation of oscillations if these factors are indeed linked.
 
In this study, Leu, Glineur and Liberati modulated expectations of pain (low or high intensity) in 40 participants prior to delivering thermal cutaneous stimulation (low, medium or high intensity). They recorded how intense participants expected the pain to be, and how intense they felt it to be, as well as EEG to assess oscillatory differences across the expectation and intensity conditions.
 
The results confirmed that there was a strong effect of expectation on the perceived stimulus intensity. However, contrary to the hypotheses, this was not reflected in the cortical oscillations. Overall this indicates a possible dissociation between perceived pain and modulation of ongoing oscillations in the theta, alpha and beta bands.
