Recommendation

Single gene alterations have been estimated to account for nearly half of neurodevelopmental conditions (NDCs), providing a crucial opportunity for animal models to understand the underlying mechanisms, causes and potential treatments. The use of systematic reviews (SRs) can, in principle, provide a powerful means to synthesise this evidence-base; however, the reporting quality of previous SRs in preclinical animal research has been found lacking (Hunniford et al., 2021). In the current study, Wilson et al. (2023) will undertake an umbrella review – a systematic review of systematic reviews – to assess the characteristics and reporting quality of SRs that, in turn, synthesise research in genetically-modified animals to model NDCs. In particular, the authors will extract key features of reviews (including, among others, the aim and primary research questions, relevant animal model, and number of studies in the SR), in addition to quality indicators such as risk of bias and completeness of reporting. In doing so, the authors aim to enhance guidance on the conduct and reporting of systematic reviews in this area.
 
The Stage 1 manuscript was evaluated over two rounds of in-depth review. Based on detailed responses to the reviewers' comments, the recommender judged that the manuscript met the Stage 1 criteria and therefore awarded in-principle acceptance (IPA).

I thank the authors for responding to my comments with such great care. I see that most of my suggestions were implemented and I am particularly happy that the PRISMA guidelines are now adhered to and PRISMA checklists are provided in the osf.io project. (Indeed, the PRISMA list for protocols that the authors used fits the purpose better than the PRISMA 2020 checklist that I had originally suggested.)

Some remaining concerns or suggestion are as follows:


1. Risk of bias: The authors state that they will not conduct a risk of bias analysis, but they actually are - risk of bias is another term used for quality assessment (which is one of the three aims of the review). So, risk of bias (with a note maybe that it is performed under the term "quality assessment"?) could be added to the checklist and the abstract. 


2. One of my main suggestions (I also included it in the first round of reviews), is that an explanation of why NDC were chosen as the  conditions that will be the focus of this review, should be given. Moreover, why are you interested in those specific 102 genes (or Rett or Fragile X)? What is their clinical importance, for example? Moreover, why did you focus on genetically modified animals and not other animal models? These choices are fine, but they need to be justified in the introduction. In their response to my comment in the previous round of reviews the authors comment that they now mention the "litter effect" in terms of NDC research, but this does not justify why they want to focus on NDC in the first place.  


3.  In the PRISMA 2015 protocol checklist it is stated by the authors that the CRediT taxonomy was used, but it was not (e.g. "critical insights, preliminary searches", etc. are not part of the CRediT taxonomy; https://credit.niso.org). I do not object with the way credit is given now, but if the authors want to state that they used the CRediT taxonomy, they should make appropriate changes to their "Contributors" section. 

 

4. Item 5c in the PRISMA 2015 protocol checklist states: "Describe roles of funder(s), sponsor(s), and/or institution(s), if any, in developing the protocol" - I believe the answer here is "none" (as the funders should have had no role in developing the protocol" and not "PhD studentship". 

 

5. Correct me if I am wrong, but I do not think preprints are included in PubMed, Embase, or Web of Science (and I think this also applies to conference abstracts). If this is the case, then where will preprints and conference abstracts be sought from?


6. Abstract: "...however, methods differ depending on the type of research being reviewed". Is this sentence necessary? I mean, will "type of research" be a variable of interest in your review?

 

7. Abstract: "We will annotate various characteristics from included systematic reviews and assess the reporting quality of each included review will be assessed using an adapted version of the PRISMA checklist (PRISMA-Pre). We will not conduct a risk of bias assessment." This sentence has a grammatical issue.


8. Abstract: "We will not conduct a meta-analysis. " I don't think this sentence is necessary (and eats away your precious word count!)


9. Minor comment: Please add a comma "," before "and" only if three or more items are listed (if you decide to use this "Oxford comma" please do it consistently). 


10. Introduction (first sentence): "De novo genetic alterations have been implicated in several neurodevelopmental conditions (NDCs), characterised by intellectual disability (ID), epilepsy, and autism spectrum condition (ASC). " Is this sentence needed? I mean, you do not focus on "de novo alterations" (in fact they are not mentioned again in the manuscript), so there is no reason to start the manuscript with this information. Do you mean genetics in general maybe?


11. Introduction (paragraph two): All these variables (that introduce error) are not an object of this review. Then why mention them in the introduction (esp. so early on)? You will not investigate bias on a study level, but on a systematic review level (and in fact only one of the many items of the PRISMA-Pre checklist, is "Report the risk of bias of the primary studies (individual studies/across outcomes)" - so, this cannot be so central in the introduction. Maybe if the introduction is restructured to first say how clinicians use systematic reviews, what systematic reviews are, how they can help identify bias and then (briefly) which are the forms of bias? In any case, the focus of this review is the quality of the systematic reviews, not the quality of the primary studies included in said reviews. I trust that the introduction might be improved if the senior authors used some of their expertise to build the rationale.


12. Introduction (paragraph two): Please explain what the "litter effect" is. 

 

13. Inclusion and exclusion criteria: "We will include systematic reviews or meta-analyses that include animal studies, either as a review limited to animal studies or ones which include them alongside other study types (e.g. clinical studies). We will exclude publications which are not systematic reviews or meta-analyses, as well as systematic reviews or meta-analyses which do not include animal studies." These two sentences say the same thing -why not say "We will only include..." and remove the second sentence? (Similarly for the second criterion, it feels like the second sentence repeats info already presented in the first sentence.)


14. Minor comment: Quite a number of typos are to be found in this version of the manuscript (e.g., abstract, "Search Strategy", second like "models" should be "model").

At the outset I would like to apologise for the delay in relaying this editorial decision. The original recommender who was handling your submission obtained two reviews quite quickly in September, and was then waiting on a third review which is now very late (and has been cancelled). At the same time, the recommender for your submission unfortunately became unavailable. As a result, I have taken over the handling of your submission and we have put in place  measures at PCI RR to prevent such delays occuring in the future.

As you will see, the reviews are critical but constructive and I believe this submission is a promising candidate for Stage 1 in-principle acceptance. Review 1 (provided by Papadatou-Pastou) is primarily concerned with the clarity of the aims and rationale, and while highly critical in places, the evaluation also provides numerous helpful recommendations for revision. Thank you for inviting me to review this registration. I have indeed reviewed the manuscript of the Stage 1 Registered Report entitled "Umbrella review of ASD animal  models: A  Stage 1  Registered  Report". First of all, let me congratulate the authors for not just pre-registering their review, but actually choosing to follow the Registered Reports path.


This review is interesting and should be a very valuable addition to the literature, once published. At the moment, though, I am afraid that the Stage 1 manuscript suffers from a number of serious flaws and needs to be rejected. The most important area for improvement is making clear what the rationale behind this review is and what it is trying to achieve.


Below you can find my comments and suggestions:


Abstract:


1. Mentioning intellectual disabilities (ID) is not needed, as this study focuses on ASD. I was actually confused while reading the abstract as to whether ID is part of the focus of this review or not. In a bit more detail, in the first sentence as well as in the phrase "summary of ASD/ID research", ID is mentioned, but in the search strategy section only ASD is mentioned. Moreover, only ASD is mentioned in the title. This is confusing to the reader as mentioned above - just by reading the title and abstract it should be clear whether ID is included in this review or not. 


2. The acronyms ASD and ID are not explained in the abstract.


3. Second line: Please replace "autism spectrum disorders" with "autism spectrum disorder" (singular). Please also replace "intellectual disabilities" with "intellectual disability". This is how they are mentioned in DSM-5.


4. "Living evidence summary" is a term reserved for the cases when the summary is regularly updated. In this case, the  review will be just published once, with no updates after it. So, this term is not appropriate. (Please correct me if I have misunderstood something here.)


5. I would suggest rewriting the abstract to take into account the PRISMA  2020 extension for Abstracts. A checklist is to be found here: https://prisma-statement.org/documents/PRISMA_2020_abstract_checklist.pdf. At the moment, a number of elements are missing from the abstract, for example inclusion and exclusion criteria as well as whether a risk of bias analysis will be conducted. I understand that there is a word limit for the abstract, but maybe the authors can fit in some of this info. They are also advised to submit the PRISMA 2020 extension for Abstracts checklist as part of the supplementary material.


Main text


Major points


1. The rationale behind this review is not clear. In the introduction there is some discussion about how systematic reviews identify areas of poor methodological quality or high risk of bias within a research literature. Is this the drive behind this review of systematic reviews? It is not clear to me how this will be achieved. Will the collected systematic reviews be then summarized with regards to the risk of bias they have identified? If so, shouldn't reporting risk of bias be one of the inclusion criteria? Or will the authors assess whether the reviews themselves have run such an analysis? It is mentioned that the authors will identify which tools will be used to measure risk of bias in the included studies, but not how their review will contribute towards identifying the risk of bias in the literature.


2. Another area that is not clear is whether literature on ID is going to be reviewed (at some point it becomes clear that the answer is no, but in the beginning of the introduction the reader is made to think that it is, as ID is mentioned in the very first sentence and then a definition of ID is given in the second sentence).


3. It should also be made clear (starting from the title) if this review concerns only genetically-modified animals or all animal models. Four types of animal models are used in preclinical research: (1) disease induction models, (2) xenograft animal models, (3) inbred strains, and (4) transgenic models, so it would be good to clarify if the focus of this review is only on the latter. 


4. An explanation (even short) of why ASD was chosen as the disease model that will be the focus of this review, should be given. 


5. The aims of this review are presented three times in the manuscript (abstract, introduction and methods), each time with a slightly different wording (minor point: the aims do not need to be repeated in the methods section). The slight change of wording is not problematic, but when the essence changes this confuses the reader. For the second aim we read "summarize the focus of these reviews" (abstract)/ "the aims of these reviews" (introduction) / "the aim of each review and what research questions does it seek to ask" (methods). These are slightly different aims. Especially with regards to "summarize the focus of these reviews" I am not sure if the authors refer to the fact that they also plan to present "a descriptive summary of data extracted from each included systematic review", as they mention in the methods section. In other words, it is no clear to me if the main aim of this review of reviews is to map systematic reviews already conducted, if it is to identify methodological issues in primary research (as mentioned in my first comment, e.g. risk of bias in the literature), or if it (further) aims to summarize the findings in the literature, by means of presenting (or even synthesizing) the findings of the included systematic reviews. 


6. On a related note, in addition to the three aims, in the methods section, a few other aims are identified: "(i) Whether the review only included animal studies, or also included clinical or in vitro studies, (ii) Which animal models the review included, (iii)The total number of studies included in the systematic review, (iv) The total number of studies investigating relevant animal models". These are listed under "systematic review aims and research question', which is not a suitable place. These are rather characteristics of the studies. Maybe the characteristics of the systematic reviews is another aim? This goes again to the fact that the objectives of this review are not clear.


7. The authors label their review an "umbrella review" (although in the methods they also refer to it as a "systematic review of systematic reviews".) However, the aim of an umbrella review is to determine what is known on a topic, what remains unknown, and to make recommendations for what requires further research. This is very different to giving a descriptive summary of data from each included systematic review -an umbrella review is about synthesizing these findings (and reaching overarching conclusions), not presenting summaries side by side. So, if the authors only want to focus on methodological issues (e.g., identify risk of bias) or to just present the number and different characteristics of published systematic reviews, this sounds more like a "scoping review of systematic reviews". Scoping reviews aim to map a research area and not to synthesize findings. However, if the authors opt for an umbrella review, then the relevant literature should be summarized in the intro.


Methods: 


1. The same criterion (animal models) is given both as an inclusion and as an exclusion criterion (with a slightly different wording, but it is the same criterion). Just mentioning it once suffices. The title of this section could even be modified to "inclusion and exclusion criteria", if the authors prefer it that way. 

2. On the same note, it is not clear to me what "Other criteria - conference abstracts and preprints" means as  an inclusion criterion. Do papers need to be conference abstracts and preprints to be included?


3 . Exclusion criteria: "Publications where we cannot access the full-text". It is the first time I come across this criterion. Genetically-modified animals are rather new (I mean a few decades) so I cannot see how one will fail to access the full text, either through their library, an inter-librayr loan or even by purchasing the full text directly. 

4. Inclusion criteria: The species of the animals should mentioned (if systematic reviews on all species will be reviewed, this should be mentioned too)

5. Exclusion criteria: "systematic reviews which do not search at least two bibliographic databases". Why is that an exclusion criterion? If one of the aims is to "identify the number of systematic reviews previously conducted", then shouldn't all systematic reviews be included? And then, one can perform a quality assessment of the included systematic reviews.


6. It was interesting to see that the authors plan to "screen studies based on full text to avoid erroneously excluding systematic reviews which do not report their inclusion criteria in their abstract" .This is fine, but -from my experience in systematic reviews and meta-analysis- a search could give back hundreds if not thousands of papers. So, the time resources needed to read each full text will not be realistic. I would suggest that the authors use a hierarchical process, by first screening titles (some papers will obviously be on a different field/irrelevant), then abstracts (or titles and abstracts together), and then the full text. Each time, one only excludes the papers they are sure do not meet the criteria and then takes the rest of the papers to the next step to see if the relevant information is there. I understand that some of the criteria might not be identifiable in the abstracts, but this is always the case in meta-analyses. This is why we never stop at the abstracts during screening. 


7. The authors should include a PRISMA 2020 checklist for their own review (on what they plan to do) and include it as supplementary material. By doing so, they can also identify a number of elements currently missing from their registration, e.g. whether they plan on conducting a risk-of-bias analysis (I understand there is discussion on biases in the manuscript, but in the methods it is not clear if this will be conducted and how). Some other elements are missing too -please check the PRISMA 2020 checklist. 


Minor
1. The phrase "will place no restriction on publication date or language" appears twice in the methods.

2. The definition given in the first sentence of the introduction applies to ASD only, but the way this sentence is written reads like it refers to all neurodevelopmental disorders (including ID, when a different definition for ID is given in the second sentence).


3. The introduction needs to be rewritten with a focus on less repetition, better flow and better justification for the need for this review. For example, paragraphs 2 & 3 & 4 could be merged. Paragraph 3 says more or less the same thing as the beginning of paragraph 4.


4. It is mentioned that the analysis code will be available online. Which analysis are the authors referring to? Reviews do not include analysis.


I understand that there are a lot of suggestions made above, but let me also say that it was great to see a detailed list of all the search terms to be used as well as a mention of the software to be used. Again, following the Registered Report route is highly commendable and I do hope a version of this work finds its way to the published literature in the future. Systematic reviews and meta-analysis are getting more and more technical, therefore there is learning curve, but I am sure the authors will get there very soon!

## Criteria

> 1A. The scientific validity of the research question(s). This criterion addresses the extent to which the research question is scientifically justifiable. Valid scientific questions usually stem from theory or mathematics, an intended or possible application, or an existing evidence base, and should be defined with sufficient precision as to be answerable through quantitative or qualitative research. They should also fall within established ethical norms. Note that subjective judgments about the importance, novelty or timeliness of a research question are NOT a relevant criterion at PCI RR and do not form part of this assessment.

I am convinved that this is a valid research question.

> 1B. The logic, rationale, and plausibility of the proposed hypotheses, as applicable. This criterion addresses the coherence and credibility of any a priori hypotheses. The inclusion of hypotheses is not required– a Stage 1 RR can instead propose estimation or measurement of phenomena without expecting a specific observation or relationship between variables. However, where hypotheses are stated, they should be stated as precisely as possible and follow directly from the research question or theory. A Stage 1 RR should also propose a hypothesis that is sufficiently conceivable as to be worthy of investigation. The complete evaluation of any preliminary research (and data) in the Stage 1 submission (see Section 2.7) is included within this criterion.

Not applicable, this is a study that measures aspects of reviews.

> 1C. The soundness and feasibility of the methodology and analysis pipeline (including statistical power analysis or alternative sampling plans where applicable). This criterion assesses the validity of the study procedures and analyses, including the presence of critical design features (e.g. internal and external validity, blinding, randomisation, rules for data inclusion and exclusion, suitability of any included pilot data) and the appropriateness of the analysis plan. For designs involving inferential statistics and hypothesis-testing, this criterion includes the rigour of the proposed sampling plan, such as a statistical power analysis or Bayesian alternative, and, where applicable, the rationale for defining any statistical priors or the smallest effect size of interest. For programmatic RRs (see Section 2.15), this criterion captures the assessment of whether the separate study components are sufficiently robust and substantive to justify separate Stage 2 outputs.

I am moderately convinced of the soundness and feasibility of the methods in this study.

I use 'moderately' as I wonder (1) how many reviews there are approximately and (2) how
long it approximately takes for a person to do one review. 
I assume this simple calculation has been done, yet I would appreciate to see
it in the paper.

> 1D. Whether the clarity and degree of methodological detail is sufficient to closely replicate the proposed study procedures and analysis pipeline and to prevent undisclosed flexibility in the procedures and analyses. This criterion assesses the extent to which the Stage 1 protocol contains sufficient detail to be reproducible and ensure protection against research bias, such as analytic overfitting or vague study procedures. In general, meeting this requirement will require the method section(s) of a Stage 1 protocol to be significantly longer and more detailed than in a regular manuscript, while also being clearly structured and accessible to readers. This criterion also covers the extent to which the protocol specifies precise and exhaustive links between the research question(s), hypotheses (where applicable), sampling plans (where applicable), analysis plans, and contingent interpretation given different outcomes. Authors are strongly encouraged to include a design summary table in their Stage 1 protocols that make these links clear (see Section 2.16 for examples). Note that in some circumstances, authors may wish to propose a more general analysis plan involving a blinded analyst rather than a precise specification of data analyses. Such submissions are acceptable and will meet this criterion provided the blinded analysis procedure is specified in reproducible detail, and provided the blinding procedure itself is sufficiently robust.

I am convinced that the methods are clear.

> 1E. Whether the authors have considered sufficient outcome-neutral conditions (e.g. absence of floor or ceiling effects; positive controls; other quality checks) for ensuring that the obtained results are able to test the stated hypotheses or answer the stated research question(s). This criterion addresses, where applicable, the extent to which the proposal pre-specifies data quality checks that will reveal whether the results are able to answer the research question(s). Such tests come in many forms and, depending on the design or subject area, are not always possible or appropriate. Where they are proposed they must be pre-specified and “outcome-neutral”, meaning that they are designed prior to knowledge of the results and are independent of the main study hypotheses. In experimental studies, such tests often consist of positive controls (also referred to in different fields as “manipulation checks” or tests of “intervention fidelity”). In observational studies they might include parallel measurements to replicate an established fact, thus confirming that the design or instrumentation is sufficiently sensitive to detect the existence of the phenomenon that forms the main focus of the research. This 1993 study which used the Galileo spacecraft to search for signs of life on Earth provides a signature example of a positive control.

I am convinced that the outcome is neutral.

## Abstract


### Objective

Research using animals to model neurodevelopmental disorders, including autism spectrum
disorders and intellectual disabilities, help us understand the underlying biology and develop
treatments and support tools. To inform the development of a living evidence summary of ASD/ID
research, we plan to conduct an umbrella review (a systematic review of systematic reviews) in this
field. This will help us (1) identify the number of systematic reviews previously conducted, (2)
summarise the focus of these reviews, and (3) assess the quality of reporting of these reviews.

### Search strategy

We will search NCBI PubMed, Ovid Embase, and Web of Science to identify systematic reviews of
animal models which use manipulations of 102 genes for which there is high confidence in a causal
relationship to ASD. We will place no restriction on publication date or language.

 

### Screening and annotation

Two independent reviewers will screen studies based on full text. Included systematic reviews will
be annotated for protocol availability, scope of the review, and inclusion criteria of the review. We
will also assess the reporting of each included review using an adapted version of the PRISMA
checklist.

### Data management and reporting

We will not conduct a meta-analysis. We will present a descriptive summary of data extracted from
each included systematic review and score each included review against the adapted PRISMA
checklist.

## Strengths and limitations of this study

 * To maximise the sensitivity (recall) of our search, we will use broad search terms related to
   ASD or neurodevelopment, in parallel with terms related to individual ASD-related genes,
   and screen studies based on full text to avoid erroneously excluding systematic reviews
   which do not report their inclusion criteria in their abstract.

 * We will use a draft extension of the PRISMA guidelines specifically designed for systematic
   reviews of animal studies, to assess the reporting quality of included systematic reviews.

 * This extension remains in draft, is not yet an official extension to PRISMA and may be subject
   to change.

## Introduction

Neurodevelopmental disorders (NDDs), such as autism spectrum disorder (ASD) and intellectual
disability (ID), are lifelong conditions characterised by difficulties with socialisation and
communication and the presence of restrictive and repetitive behaviours and interests. Intellectual
disability is characterised by significantly impaired intellect and is defined by an IQ of less than 70.

Animals can be used to model the genetic causes of NDDs and their associated cellular, circuit and
behavioural traits. Such research plays a vital role in developing our understanding of the biology
underlying NDDs (basic research) and in the testing potential drug treatments (preclinical research).

Researchers have developed numerous genetically modified animal models to study ASD. Such
models allow researchers to understand the function of ASD-related genes during brain
development and allow for the development and testing of therapeutics prior to clinical trials.

However, despite extensive research, successful translation of laboratory findings to the clinic is rare
(Pankevich et al., 2013; Silverman et al., 2022). Many factors may contribute to this lack of success
including target choice, outcome measures insensitive to change, disease stage at which treatment
is initiated, lack of construct validity and high risk of bias in the preclinical 
literature [richelbilderbeek: describe bias here, not below].

The successful translation of laboratory findings to clinical trials is a major challenge in many areas of
biomedical research (Ioannidis, 2006), and is not limited to neurodevelopmental research. Poor
methodological quality and high risks of bias – such as lack of randomisation of animals or lack of
blinded outcome assessment – can also contribute to this translational barrier by compromising the
internal validity of a study (Krauth et al., 2013). Bias describes a systematic error within a study,
caused by a variable other than the intervention of interest, that influences the measured outcome [richelbilderbeek: describe bias above, not here].

By ensuring that a study is internally valid, we can be more confident that – apart from random error
– findings from the study reflect a true cause-and-effect relationship between the intervention of
interest and the measured outcome in that experimental setting.

Systematic review provides a well-developed method for identifying areas of poor methodological
quality or high risk of bias within a research literature. Systematic review is a research method used
to summarise and appraise all available evidence related to a pre-specified topic (Egger et al., 2001),
and can identify areas for improvements which might increase internal validity and research rigour
and reproducibility. Clinicians have used systematic reviews to inform evidence-based healthcare since the 1980s.

Findings from such reviews have led to considerable improvements in the way clinical trials are
conducted and reported (Plint et al., 2006). Over the last two decades, researchers have adopted
systematic review methods to summarise and appraise evidence from laboratory animal studies (de
Vries et al., 2014) and have achieved similar success in research improvement (McCann et al., 2016;
Ramirez et al., 2017).

However, the fundamental differences [richelbilderbeek: which fundamental differences?

between clinical and animal studies means that systematic
review methodologies must be appropriately adapted. The development of several tools has been
instrumental in supporting researchers conducting systematic reviews and meta-analyses of animal
studies, including checklists for assessing the risk of bias (Hooijmans et al., 2014) and reporting
quality (Macleod et al., 2004) of laboratory animal studies, and a protocol template for systematic
reviews of animal intervention studies (de Vries et al., 2015). Despite this, reporting quality of animal
systematic reviews is low (Mueller et al., 2014; Hunniford et al., 2021).

This umbrella review aims to understand (1) how many systematic reviews have been conducted in
the area of ASD animal research, (2) the aims of these reviews, and (3) the reporting quality of these
reviews. As a secondary output, this research will help inform the creation of a living evidence
summary (Hair et al., 2022) [richelbilderbeek: I feel the reference to Hair et al., 2022

of animal research modelling ASD which might inform future systematic
reviews, by ensuring that such a tool is fit for purpose and provides the appropriate resources to
allow researchers to conduct high quality systematic reviews. We chose to focus on systematic
reviews summarising research using genetic animal models of any of the 102 ASD-related genes
identified through large-scale exome sequencing by Satterstrom et al. (2020), as these are all genes
where there is high confidence in their clinical relevance [richelbilderbeek: reference?].

## Methods

To date, we have completed preliminary searches and optimised our search strategy to inform the
development of this protocol.

## Research question

This systematic review of systematic reviews seeks to investigate (1) how many systematic reviews
have been conducted in this area, (2) what is the aim of each review and what research question
does it seek to ask, and (3) and what is the quality of reporting of these reviews.

## Bibliographic search

We will conduct systematic literature search on three electronic databases: PubMed (accessed via
NCBI), Embase (accessed via Ovid), and Web of Science Core Collection.

Our search strategy includes three components: (1) broad terms related to autism,
neurodevelopment, or ASD-related genes; (2) terms related to animal models, and (3) terms related
systematic reviews or meta-analyses. Terms used to identify animal models were taken from van der
Mierden et al. (2022) and terms used to identify systematic reviews and meta-analyses were taken
from Langendam et al. (2021). Full search terms are given in Appendix 1.

We will not restrict searches by publication date or language. If we include studies without an
English language full-text, we will use Google Translate to translate the full-text into English. Where
citations appear in multiple databases, we will remove duplicate versions of the citation using the
ASySD tool (Hair et al., 2021).

## Screening

We will upload our search results, with duplicate citations removed, to the Systematic Review
Facility (SyRF) platform (Bahor et al., 2021) for screening. Two independent reviewers will screen
each publication for inclusion and any disagreements will be reconciled by a third independent
reviewer. We plan one round of screening, where we will screen the full-texts of all studies retrieved
from our searches against our inclusion and exclusion criteria, to avoid potentially excluding
systematic reviews where the decision for inclusion rests on information contained in the full text
but not in the abstract (Wilson et al., 2022).

We will retrieve full text PDF files using the automated find full text feature in EndNote 20 or via
handsearching. If we cannot access the full text using our institutional subscriptions, we will request
the full text via interlibrary loan.


### Inclusion and exclusion criteria

Studies will be screened according to the criteria outlined below:

 * Inclusion criteria:
   * Study design – systematic reviews or meta-analyses that include animal studies,
     either as a review limited to animal studies or one which includes them alongside
     other study types.
   * Animal models – systematic reviews or meta-analyses which include studies
     investigating a genetic animal model based on at least one of the 102 ASD-related
     genes identified by Satterstrom et al. (2020).
   * Other criteria – conference abstracts and preprints.

 * Exclusion criteria:
   * Study design – systematic reviews of meta-analyses which do not include animal
     studies, or systematic reviews which do not search at least two bibliographic
     databases [richelbilderbeek: I would enjoy to hear why this is an exclusion criterium]. Other publication types such as non-systematic reviews, letters,
     editorials, or opinion pieces.
   * Animal models – systematic reviews or meta-analyses which do not include animal
     models, or where the animals are not models of at least one of the 102 genes
     identified by Satterstrom et al., (2020).
   * Other criteria – publications where we cannot access the full-text.

We will place no restriction on publication date or language.

## Data extraction

Two independent reviewers will conduct data extraction. Discrepancies between reviewer decisions
will be reconciled by a third independent reviewer. We will carry out data extraction using the SyRF
platform, and will collect the following information:
 * Bibliographic data
 * Systematic review aims and research question
 * Reporting quality


## Bibliographic data

We will extract names of first authors, year of publication, title, and DOI of each included study.

### Systematic review aims and research question

To understand the purpose and scope of each included systematic review, we will extract the aim of
each systematic review and the primary research question each review seeks to ask. Additionally, we
will extract:

 * Whether the review only included animal studies, or also included clinical or in vitro studies
 * Which animal models the review included
 * The total number of studies included in the systematic review
 * The total number of studies investigating relevant animal models


## Reporting quality

We will assess the reporting quality of each included systematic review using the 46-point checklist developed by Hunniford et al. (2021). The checklist is adapted from the PRISMA guidelines for
systematic reviews and is more specific to systematic review of animal studies. However, the
checklist is not currently an official extension to PRISMA. Although it is not recommended to use the
general PRISMA guidelines to assess in vivo systematic review reporting 
quality [richelbilderbeek: I would enjoy a reference to this statement :-) ], the adapted
checklist for preclinical systematic reviews has been designed for this purpose.

The checklist items are written in full in Appendix 2.

## Data synthesis

We will not conduct a meta-analysis. However, we will present a descriptive summary of data
extracted from each included systematic review.

We will present bibliographic details of each included systematic review and provide a narrative
review of each review’s research question and aim [richelbilderbeek: I enjoy the enthusiasm
here, but I see no reason why to provide this narrative. I would enjoy to see why this narrative
is provided, especially as the checklist already contain quite some 'describe X' items]. 
We will then score each included systematic
review using the PRISMA modified reporting checklist and provide a summary table detailing which
items of the checklist each review met. 
Additionally, the checklist will allow us to assess which tools
are currently being used to conduct systematic reviews in this field, including the tools used to
screen studies (Checklist Item 6a), extract numerical data (Checklist Item 17a), and measure study
quality or risk of bias (Checklist Item 19).
